Comparative Histochemical Studies on the Mucosubstances and Some Enzymes in the Small Intestine

Abstract
Comparative histochemical studies were done on the mucosubstances in the goblet cells and Brunner¢¥ s glands and on the distribution of alkaline phosphatase and acid phosphatase in the epithelium of the villi and crypts, in the small intestine of the mouse, rat, guinea pig, rabbit, cat, doo and human fetus. The mucosubstances were observed with alcian blue(pH 2. 5), PAS and alcian blue(pH 2. 5)-PAS stainings and the alkaline and acid phosphatase were observed with Gomori¢¥s modified methods.
The results obtained were as follows:
1. The mucosubstances in the goblet cells of the small intestine of human fetus and animals were composed of acid and neutral mucosubstance.
2. The proportion and location of both mucosubstances in the goblet cells were different within each animal. That is, in the villi of the small intestine of the mouse, and in the small intestine of the human fetus and dog were more neutral, and those in the crypts of the small intestine of the mouse, in the small intestine of the rabbit and guinea pig and in the duodenum of the cat were more acidic. In the small intestine of the rat and in the jejunum and ileum of the cat there were nearly equal amount of neutral and acid mucosubstances.
3. The mucosubstance in the Brunner¢¥s glands of the guinea pig and rabbit were composed of neutral and acid mucosubstances but those of the other animals and human fetus were composed of only neutral.
4. The alkaline phosphatase activity in the epithelium of the villi in the small intestine appeared moderately to strongly positive in the human fetus and animals, but that of the crypts in the small intestine appeared differented in the neck and base of the crypts. That is, the alkaline phosphatase activity in the neck of the crypts appeared moderately to strongly positive in the human fetus and animals except the rabbit, which appeared weekly positive, but that in the base of the crypts appeared almost negative in the human fetus and animals except the cat, which appeared moderately positive.
5. The acid phosphatase activity in the epithelium of the villi in the small intestine appeared strongly positive in all the materials except the rabbit and human fetus, these appeared almost negative, and that of the crypts in the small intestine appeared moderately to strongly positive in the cat and weekly to moderately positive in the guinea pig, but almost negative in the other animals and human fetus.
